چکیده

The extent of angiogenesis and/or vascular endothelial growth factor (VEGF) expression in neuroblastoma tumors correlates with metastases, N-myc amplification, and poor clinical outcome. Recently, we have shown that insulin-like growth factor-I and serum-derived growth factors stimulate VEGF expression in neuroblastoma cells via induction of hypoxiainducible factor-1A (HIF-1A). Because another marker of poor prognosis in neuroblastoma tumors is high expression of brain-derived neurotrophic factor (BDNF) and its tyrosine kinase receptor, TrkB, we sought to evaluate the involvement of BDNF and TrkB in the regulation of VEGF expression. VEGF mRNA levels in neuroblastoma cells cultured in serum-free media increased after 8 to 16 hours in BDNF. BDNF induced increases in VEGF and HIF-1A protein, whereas HIF-1B levels were unaffected. BDNF induced a 2to 4-fold increase in VEGF promoter activity, which could be abrogated if the hypoxia response element in the VEGF promoter was mutated. Transfection of HIF-1A small interfering RNA blocked BDNFstimulated increases in VEGF promoter activity and VEGF protein expression. The BDNF-stimulated increases in HIF-1A and VEGF expression required TrkB tyrosine kinase activity and were completely blocked by inhibitors of phosphatidylinositol 3-kinase (PI3K) and mammalian target of rapamycin (mTOR) pathways. These data indicate that BDNF plays a role in regulating VEGF levels in neuroblastoma cells and that targeted therapies to BDNF/TrkB, PI3K, mTOR signal transduction pathways, and/or HIF-1A have the potential to inhibit VEGF expression and limit neuroblastoma tumor growth. Tropomyosin-related kinase B mediated signaling contributes to the induction of malignant phenotype of gallbladder cancer

This study aims to demonstrate the clinical and biological significance of Brain derived neurotrophic factor (BDNF)/Tropomyosin-related kinase B (TrkB) signaling in gallbladder cancer (GBC) through a series of in vitro and in vivo experiments. TrkB expression was detected in 63 (91.3%) out of 69 surgically resected primary GBC specimens by immunohistochemistry. TrkB expression in the invasive f...

Effect of 8 weeks of Aerobic Training on Genes Expression of Hypoxia Inducible Factor HIF-1α, Vascular Endothelial Growth Factor (VEGF) and Angiostatin in Hippocampus of Male Rats with Wistar Model

Introduction: Many studies have been done about the effects of exercise on angiogenic inhibitor and stimulator factors in muscles, but few studies have examined the role of these factors in the brain especially the hippocampus. Therefore, the purpose of the current study was to investigate the effect of 8 weeks of aerobic training on gene expression of HIF-1α, VEGF and angiostatin in hippocampu...

Blockade of Hypoxia: The Impact on Tumor Growth in an Experimental Tumor Model

Background: Tumor microenvironment is an active factor participating in immunoregulation, thereby preventing immunosurveillance and limiting the efficacy of anticancer therapies. Hypoxia as a major characteristic of solid tumors causes the expression of Hypoxia-Inducible Factor-1α (HIF-1α). This is a transcription factor that mediates hypoxic responses of tumor cells and involves in the express...
